The earliest study on the anti forgetting potential of pracetam is reported. The forgetting reversal potential of prasidan reaches the effective peak (96% reversion) at 50mg/kg (effective range is 12.5-80mg/kg, intraperitoneal injection). The results show that the effect of prasidan is greater than piracetam, and prasidan is the basis of prasidan synthesis.
In a study using prasidan as a comparison to assess the effect of urinary acid (NMDA agonist at glycine binding site) in dogs, prasidan of 30mg/kg can enhance object recognition and memory. Improvements in object recognition have been noted elsewhere, with 30mg/kg prasidan (more effective than 15mg/kg and 60mg/kg) and 400mg/kg piracetam. The antihypertensive contraceptive effect was also improved when the mice were given pracetam (100mg/kg) 1-2 hours before the test. The efficacy was similar to that of olacetam (the same dose), but better than piracetam, and the same or excellent performance as alacetam. One study found that performance in the water maze also improved, with a lower 10mg/kg and a higher dose more effective.
The use of 7.5-15mg/kg prasidan in rats after 7 weeks training in 16 arm radial maze task can improve the reference (long-term) memory, but not significantly affect the working memory. Although the trend of 15mg/kg is more effective than 7.5mg/kg, the difference seems not significant.
